用慢性日本血吸虫病病人血清筛选以βgill构建的日本血吸虫中国大陆株成虫cDNA表达库,获得一阳性克隆Sjgt4,将其插入片段经聚合酶链反应(PCR)法扩增后亚克隆入表达载体pTrcHisB并进行表达,获37kDa融合蛋白,它可溶于8M尿素,并可被日本血吸虫病病人血清中特异抗体所识别。以此37kDa融合蛋白免疫家免所得的抗血清,可特异地识别日本血吸虫成虫抗原的67kDa分子。
The cDNA library of adult Schistosoma japonicum strains with βgill was screened from serum of patients with chronic schistosomiasis japonica and a positive clone Sjgt4 was obtained. The inserted fragment was amplified by polymerase chain reaction (PCR) and subcloned into the expression vector pTrcHisB And expressed, get 37kDa fusion protein, which is soluble in 8M urea and can be recognized by specific antibodies in the serum of patients with schistosomiasis japonica. The 37kDa fusion protein was used to immunize the home-made antisera to specifically identify the 67kDa antigen of adult schistosoma japonicum antigen.
