目的探讨白介素6(IL-6)对卵巢癌SKOV3细胞株中B-细胞淋巴瘤2(Bcl-2)、细胞周期蛋白D1(CyclinD1)和血管内皮生长因子(VEGF)的影响。方法不同浓度IL-6作用于细胞,找出IL-6最佳作用浓度。在IL-6最佳浓度基础上,加不同浓度的AG490。MTT、流式细胞术、RT-PCR、Western blotting检测细胞增殖、凋亡及细胞Bcl-2、CyclinD1、VEGF mRNA和蛋白表达。结果 IL-6(0、10、20 ng/mL)对细胞的作用呈浓度依赖性,促进细胞增殖,抑制其凋亡(P<0.01);Bcl-2、CyclinD1、VEGF蛋白和mRNA表达增加(P<0.01),48 h与24 h差异有统计学意义(P<0.01)。AG490(0、20、40、80μmol/L)对细胞的作用呈浓度依赖性,抑制细胞增殖,促进其凋亡(P<0.01);Bcl-2、CyclinD1、VEGF蛋白和mRNA表达下调(P<0.01),48 h与24 h差异无统计学意义(P>0.05)。结论 IL-6通过调节STAT3通路,调节细胞Bcl-2、CyclinD1和VEGF的表达,从而促进卵巢癌细胞的增殖,抑制其凋亡。
Objective To investigate the effect of interleukin-6 (IL-6) on the expression of Bcl-2, CyclinD1 and VEGF in ovarian cancer cell line SKOV3. Methods Different concentrations of IL-6 were applied to cells to find out the best concentration of IL-6. In the best concentration of IL-6, add different concentrations of AG490. MTT, flow cytometry, RT-PCR, Western blotting were used to detect the cell proliferation, apoptosis and the expression of Bcl-2, CyclinD1, VEGF mRNA and protein. Results The effect of IL-6 (0, 10, 20 ng / mL) on cells was concentration-dependent, promoted cell proliferation and inhibited apoptosis (P <0.01), and increased expressions of Bcl-2, CyclinD1 and VEGF P <0.01). There was a significant difference between 48 h and 24 h (P <0.01). The effect of AG490 (0, 20, 40, 80μmol / L) on cells was concentration dependent, inhibited cell proliferation and promoted apoptosis (P <0.01), while downregulated the expression of Bcl- 0.01), but no significant difference between 48 h and 24 h (P> 0.05). Conclusion IL-6 regulates the expression of Bcl-2, CyclinD1 and VEGF by regulating STAT3 pathway, which promotes the proliferation and inhibits the apoptosis of ovarian cancer cells.
