PA(PksmlnosenActivator)是体内一重要蛋白水解酶系统.包括tPA和uPA二型,主要水解纤溶酶原为纤溶酶,从而启动对多种细胞外基质蛋白的水解,同时还可直接裂解纤粘素、胶原酶IV和肝细胞生长因子等。PA的抑制剂PAI(PlasminogenActivatorlrl-hibitor)包括PAI-1和PAI-2两种,二者均抑制tPAuPA。PAI-1合成于角既细胞(kerationcvtKC)和其他多种细胞分布较广。PAI—2合成于角朊细胞和巨噬细胞,主要见于表皮,在妊娠期胎盘和血浆中含量明显增高。作者对tPA/PAI-2的研究工作发现,在分化中,角朊细胞产生tPA,后者破坏细胞与细胞之间的连接,介导粒层细胞中大块蛋白降解,产生一些具有化学趋化性的蛋白水解片断,核崩解;tPA还可帮助角阮细胞成层排列,并分离上皮表面已终未分化的鳞片。?*1-2则主要合成于基底上层角既细胞,沿细胞周围分布并和细胞外基质蛋白结合,调控表皮分化中tPA的复杂水解过程。最近我们发现,tPA信号仅出现在毛囊下段的基质细胞中,并选择性的表达在某特定的毛发周期中。由上可见,tPA/PAI-2系统的研究对阐明表皮细胞分化及其调控机理、细胞粘附、游走等重大理论课题有很重要的意义,对组织损伤修复过程和某些表应分化不全疾病(如牛皮癣)的病因研究等课题也有重要价值
