目的:通过比较不同侵袭能力的前列腺癌细胞netrin-1/UNC5B表达量的差异,为前列腺癌寻找新的生物标志物及治疗手段。方法:选取前列腺癌细胞系DU145、22RV1、PC3、PC3M、RWPE-1,通过RT-PCR、Western印迹检测netrin-1/UNC5B的表达及其差异,并通过免疫荧光检测配体netrin-1及其受体UNC5B在前列腺癌细胞中的定位。结果:netrin-1/UNC5B在前列腺癌细胞中均表达,侵袭能力强的前列腺癌细胞中netrin-1表达量明显增多(P<0.05),而UNC5B在RWPE-1(正常细胞)中表达量明显增多(P<0.05)。结论:netrin-1/UNC5B表达量与前列腺癌的浸润及进展程度紧密关联,有望作为一种潜在的生物标志物来预测前列腺癌的恶性程度。
OBJECTIVE: To search for new biomarkers and therapeutic approaches for prostate cancer by comparing the expression levels of netrin-1 / UNC5B in different invasiveness of prostate cancer cells. METHODS: The expression of netrin-1 / UNC5B and its difference were detected by RT-PCR and Western blotting, respectively. DU145, 22RV1, PC3, PC3M and RWPE-1 were detected by immunofluorescence. Location of Receptor UNC5B in Prostate Cancer Cells. Results: The expression of netrin-1 / UNC5B in prostate cancer cells was significantly increased (P <0.05), while the expression level of UNC5B in RWPE-1 (normal cells) was significantly higher Increased (P <0.05). Conclusion: The expression of netrin-1 / UNC5B is closely related to the degree of invasion and progression of prostate cancer, which is expected to predict the malignancy of prostate cancer as a potential biomarker.
