目的:观察利胆止痛胶囊的利胆、退黄作用和对大鼠肝损伤的保护作用。方法:采用大鼠胆管引流法观测利胆止痛胶囊对大鼠胆汁量的影响。用异硫氰酸-1-萘酯(alpha-naphthyl isothiocyanate,ANIT)造成大鼠黄疸模型,测定血清中谷丙转氨酶(al-anine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)和总胆红素(total bilirubin,TBIL)的含量。制备大鼠四氯化碳肝损伤模型,观察利胆止痛胶囊对肝损伤大鼠血清ALT、AST、碱性磷酸酶(alkaline phosphatase,ALP)、总蛋白(total protein,TP)、白蛋白(Albumin,ALB)的影响,同时观察肝脏病理学改变。用RM6240BD型多道生理信号采集处理系统记录豚鼠离体胆囊平滑肌条的张力。结果:0.36、0.72 g生药/kg利胆止痛胶囊明显增加正常大鼠的胆汁流量。利胆止痛胶囊呈浓度相关性增加豚鼠离体胆囊平滑肌条的张力,0.5g生药/L浓度则无明显影响。0.36、0.72g生药/kg利胆止痛胶囊显著降低TBIL水平(与模型组比较)。0.72g/kg的利胆止痛胶囊可降低ALT和AST。0.72生药g/kg的利胆止痛胶囊明显降低四氯化碳肝损伤大鼠血清ALT、AST、ALP水平,0.18~0.72g生药/kg的利胆止痛胶囊对TP和ALB均无明显影响;病理组织学观测显示,与正常组比较,模型组肝小叶结构破坏,肝细胞明显变性坏死和炎性细胞浸润,细胞结构凌乱。中、高剂量的利胆止痛胶囊可改善肝脏损伤,可见部分肝细胞变性坏死和炎性细胞浸润。结论:利胆止痛胶囊具有利胆、退黄和保肝降酶作用。
Objective: To observe the gallbladder, jaundice and protective effect of Lilezhitong capsule on rat liver injury. Methods: The bile duct drainage method was used to observe the effect of Lilengzhitong Capsule on the amount of bile in rats. The rat jaundice model was induced by alpha-naphthyl isothiocyanate (ANIT) and the levels of serum ALT, AST and total bilirubin Total bilirubin (TBIL) content. The model of CCl 4 -induced liver injury was established in rats. The effects of Lilingzhitong Capsule on serum ALT, AST, alkaline phosphatase (ALP), total protein (TP), albumin , ALB), while observing the liver pathological changes. Multi-channel physiological signal acquisition and processing system RM6240BD was used to record the tension of guinea pig gallbladder smooth muscle strips. Results: 0.36,0.72 g crude drug / kg Lilengzhitong Capsule significantly increased bile flow in normal rats. Liguanzhitong capsule showed a concentration-related increase in guinea pig gallbladder smooth muscle tension, 0.5g crude drug / L concentration had no significant effect. 0.36, 0.72g crude drug / kg Li Danzhitong capsule significantly reduced TBIL levels (compared with the model group). 0.72g / kg of Lilengzhitong capsule can reduce ALT and AST. 0.72 crude drug g / kg of LILE group significantly reduce serum levels of ALT, AST and ALP in CCl 4 -induced liver injury, 0.18 ~ 0.72g crude drug / kg of LILE in patients with TP and ALB had no significant effect; Histological observation showed that compared with the normal group, the model group hepatic lobule structure destruction, hepatocellular degeneration and necrosis and inflammatory cell infiltration, cell structure messy. Medium and high doses of gallbladder analgesic capsule can improve liver injury, we can see part of the degeneration and necrosis of liver cells and inflammatory cell infiltration. Conclusion: Lilengzhitong capsule has the functions of gallbladder, rehmanniae and hepatic descending enzyme.
