目的探讨宫颈癌患者行子宫动脉介入化疗联合栓塞治疗前、后宫颈癌组织miR-143和岩藻糖基转移酶4(fucosyltransferase 4,FUT4)表达变化及意义。方法 20例宫颈癌患者,采用多西他赛+顺铂方案子宫动脉介入化疗联合栓塞治疗3周后行手术治疗,采用实时荧光定量PCR法检测化疗前、后宫颈癌组织中miR-143表达,采用Western blot法检测FUT4蛋白表达,Pearson线性相关分析宫颈癌组织miR-143与FUT4表达的相关性。结果化疗后宫颈癌组织miR-143相对表达量(2.11±0.17)较化疗前(1.04±0.10)增高,FUT4相对表达量(0.38±0.08)较化疗前(0.93±0.09)降低(P<0.05);miR-143表达在化疗前、后与FUT4均呈负相关(r=-0.509,P=0.020;r=-0.563,P=0.010)。结论多西他赛+顺铂方案子宫动脉介入化疗联合栓塞治疗可上调miR-143及下调FUT4在宫颈癌组织中的表达。
Objective To investigate the expression and significance of miR-143 and fucosyltransferase 4 (FUT4) in patients with cervical cancer before and after uterine artery interventional chemotherapy combined with embolization. Methods Twenty patients with cervical cancer were treated with docetaxel plus cisplatin for 3 weeks after interventional chemotherapy and uterine artery embolization. The expression of miR-143 in cervical cancer tissues before and after chemotherapy was detected by real-time fluorescence quantitative PCR. blot assay was used to detect the expression of FUT4 protein, Pearson linear correlation analysis of cervical cancer tissue miR-143 and FUT4 expression. Results The relative expression level of miR-143 in cervical cancer tissues after chemotherapy (2.11 ± 0.17) was significantly higher than that before chemotherapy (1.04 ± 0.10), and the relative expression level of FUT4 (0.38 ± 0.08) was lower than that before chemotherapy (0.93 ± 0.09) The expression of miR-143 was negatively correlated with FUT4 before and after chemotherapy (r = -0.509, P = 0.020; r = -0.563, P = 0.010). Conclusions Docetaxel plus cisplatin can increase the expression of miR-143 and down-regulate the expression of FUT4 in uterine cervix carcinoma by uterine artery interventional chemotherapy combined with embolization.
