本实验对体外培养的新生大鼠心房和心室肌细胞进行了光镜和电镜观察,并以免疫组织化学方法对心肌细胞内心钠素的定位和含量进行了探讨。结果表明,培养的大鼠心房和心室肌细胞胞浆内均可见到心钠素免疫反应阳性颗粒,但心室肌细胞内心钠素含量很少;并观察到有含和不含心钠素阳性颗粒的两种心房肌细胞。实验证实,心钠素在培养新生大鼠心肌细胞中的定位与含量与在体时基本一致。本方法为从细胞水平研究各种因素对心钠素释放的影响提供了新途径。
In this study, the atrial and ventricular myocytes of neonatal rats cultured in vitro were observed under light microscope and electron microscope, and the localization and content of atrial natriuretic peptide (APN) in myocardial cells were studied by immunohistochemistry. The results showed that the atrial natriuretic peptide positive particles were found in the atrial and ventricular myocytes of the cultured rat hearts, but the content of atrial natriuretic peptide was very low in the ventricular myocytes; and the presence and absence of atrial natriuretic peptide Of two kinds of atrial myocytes. Experiments confirmed that atrial natriuretic cells in neonatal rat cardiomyocytes in the positioning and content of the body is basically the same. This method provides a new way to study the effects of various factors on the release of atrial natriuretic peptide at the cellular level.
