在葡萄试管苗培养过程中,先高后低的变温条件对幼苗的生长发育有利。28℃～30℃有利于根的形成,26℃～28℃是形成壮苗的适宜温度。在幼苗生长阶段光照强度应不低于1800lux。培养基中激素配比以基本培养基+IAA 0.3mg/升+IBA0.1mg/升对诱导外植体早生根并促进茎叶生长的效果最显著。影响试管苗移栽成活率的几种基质处理中以水培和砂培的效果最好。
In vitro culture of grape seedlings, the first high and low temperature conditions conducive to the growth and development of seedlings. 28 ℃ ~ 30 ℃ is conducive to the formation of roots, 26 ℃ ~ 28 ℃ is the appropriate temperature to form strong seedlings. Light intensity in seedling growth stage should be not less than 1800lux. The best proportion of hormone in the medium was that the basic medium + IAA 0.3 mg / L + IBA 0.1 mg / L had the most significant effect on inducing the root growth of explants and promoting the growth of shoots and leaves. The effects of hydroponics and sand culture were the best in several kinds of substrate treatments that affected the survival rate of transplanted in vitro plantlets.
