目的:探讨蜂蜡素对HepG2细胞HMG-CoA还原酶mRNA表达及蛋白质水平的影响。方法:采用MTT比色法,Western Blot,RT-PCR等技术方法,观察蜂蜡素对HepG2细胞存活能力、HMG-CoA还原酶mRNA表达及蛋白质水平的影响。结果:当蜂蜡素浓度为100mg·L-1或以下时,对细胞存活能力无影响,为本研究可选择的剂量范围;蜂蜡素处理HepG2细胞,随着蜂蜡素浓度升高(低、中、高浓度为0.8,8,80mg·L-1),HMG-CoA还原酶蛋白质水平和mRNA的表达均逐渐降低,呈现明显的浓度依赖关系。结论:蜂蜡素可抑制HepG2细胞HMG-CoA还原酶蛋白质及mRNA的表达。
Objective: To investigate the effect of beeswax on HMG-CoA reductase mRNA expression and protein level in HepG2 cells. Methods: The effects of beeswax on HepG2 cell viability, HMG-CoA reductase mRNA expression and protein level were observed by MTT assay, Western Blot, RT-PCR and other techniques. RESULTS: When the concentration of beeswax was 100 mg·L-1 or less, there was no effect on the viability of the cells, which was the range of doses available for this study; beeswax treated HepG2 cells with increasing concentrations of beeswaxine (low, medium, At high concentrations of 0.8,8,80 mg·L-1, HMG-CoA reductase protein levels and mRNA expression were gradually decreased, showing a significant concentration-dependent relationship. CONCLUSION: The beeswax can inhibit the expression of HMG-CoA reductase protein and mRNA in HepG2 cells.
