目的:优选白花蛇舌草的水提醇沉多糖提取工艺条件。方法:以白花蛇舌草多糖含量为指标,蒽酮比色法测定指标成分含量,在单因素试验基础上,采用L9(34)正交试验考察料液比、超声提取时间、重复提取次数和乙醇浓度对白花蛇舌草多糖含量的影响,优化其提取工艺。并用DPPH自由基清除率法对其抗氧化活性进行测定。结果:白花蛇舌草多糖最佳提取的工艺条件为料液比1:14,提取时间30~40 min,重复提取次数3次,提取乙醇浓度60%。自由基清除实验显示,白花蛇舌草多糖具有浓度相关的自由基清除能力和抗氧化能力。结论:优选的水提醇沉提取工艺稳定可行,所提取白花蛇舌草多糖具有较强的自由基清除效果,是一种极具开发潜力的优良天然抗氧化剂。
OBJECTIVE: To optimize the extraction conditions of water extract and alcohol polysaccharide from Hedyotis diffusa. Methods: The content of polysaccharides in Hedyotic diffusa was determined by anthrone colorimetric method. Based on the single factor test, the ratio of material to liquid, ultrasonic extraction time and the number of repeated extraction were determined by L9 (34) orthogonal test Ethanol concentration on the content of polysaccharide Hedyotis diffusa, to optimize its extraction process. Antioxidant activity was measured by DPPH radical scavenging assay. Results: The optimal extraction conditions of polysaccharide from Hedyotic diffusa were polysaccharide ratio 1:14, extraction time 30 ~ 40 min, extraction times 3 times and ethanol concentration 60%. Free radical scavenging experiments showed that the Hedyotis diffusa will concentration-dependent free radical scavenging and antioxidant capacity. CONCLUSION: The optimal extraction process of ethanol extraction by water extraction is stable and feasible. The extracted HTPA polysaccharide has strong free radical scavenging activity and is an excellent natural antioxidant with great development potential.
