慢性房颤犬脂肪垫Notch-1表达变化

目的 基于慢性起搏犬左心房致房颤模型的建立,研究慢性房颤犬窦房结脂肪垫(SAN-FP)神经丛中Notch-1蛋白表达变化及Notch-1、NICD含量变化,阐明SAN-FP神经丛Notch信号通路与房颤的发生发展有密切关系.方法 健康比格犬体重18±2.66 kg,年龄7-9岁.实验动物随机分为2组,假手术组(n=5)和房颤组(n=5).结果 房颤组犬经快速起搏左心耳均可成功诱发房颤.SAN-FP可见脂肪细胞中散在分布的神经丛,与假手术组比较,房颤组Notch-1蛋白表达显著增加,差异有统计学意义(P0.05).实验8周后,与假手术组相比,房颤组Notch-1和NICD浓度显著增加,差异有统计学意义(P<0.05).结论 房颤发生发展过程中SAN-FP神经丛内Notch-1可能在房颤发生的不同时期表达,Notch-1与配体结合后以NICD形式活化进而影响下游靶基因.
