目的建立高效液相色谱法测定蒙药孟根乌森乌日勒中乌头碱、次乌头碱、新乌头碱、苯甲酰乌头原碱、苯甲酰次乌头原碱及苯甲酰新乌头原碱的含量。方法哈勒C18色谱柱,以乙腈-四氢呋喃(25∶15)为流动相A,以0.1 mol·L-1醋酸铵溶液为流动相B,梯度洗脱,流速0.8 m L·min-1;柱温30℃;检测波长235 nm。结果乌头碱、次乌头碱、新乌头碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、苯甲酰新乌头原碱的浓度分别在3.48×10-3~1.74×10-1,3.24×10-3~1.62×10-1,6.96×10-3~3.48×10-1,2.06×10-3~1.03×10-1,4.02×10-3~2.02×10-1,6.66×10-3~3.33×10-1mg·m L-1(r分别为0.999 5,0.999 0,0.999 0,0.999 9,0.999 9,0.999 9)呈良好的线性关系,平均回收率分别为95.54%,103.18%,102.75%,92.29%,99.41%,107.8%,RSD分别为3.70%,1.89%,4.94%,3.67%,4.29%,3.89%。结论本方法简便、准确、重复性好,可用于孟根乌森乌日勒中单酯型生物碱和双酯型生物碱的含量测定。
