为了进一步研究rhTGF-β1抑制人肝癌细胞SMMC-7721增殖的作用机理,应用光镜、电镜观察rhTGF-β1抑制人肝癌细胞SMMC-7721增殖的形态学改变。结果显示:经rhTGF-β110ng/ml作用96小时后,SMMC-7721细胞出现曲型的凋亡形态。相差显微镜下表现为细胞变圆、体积缩小;HE染色可见核固缩;荧光显微镜观察凋亡细胞多为核亮染呈致密斑块;电镜下可见由膜包裹内容物的凋亡小体。结果证实:rhTGF-β1具有诱发肝癌人细胞SMMC-7721凋亡的作用
To further study the mechanism of rhTGF-β1 inhibiting the proliferation of human hepatocellular carcinoma cells SMMC-7721, the morphological changes of proliferation of human hepatoma cells SMMC-7721 were observed by light and electron microscopy. The results showed that after 96 hours of rhTGF-β110ng/ml, SMMC-7721 cells showed a curved apoptotic morphology. The phase contrast microscope showed that the cells became round and the volume was reduced; HE staining showed nuclear condensation; fluorescence microscopy showed that the apoptotic cells were mostly nuclear bright and dense patches; electron microscopy showed apoptotic bodies wrapped by the membrane. The results confirmed that rhTGF-β1 can induce apoptosis of hepatoma cells SMMC-7721.
