目的探讨癌基因表达的变化与辐射诱导肿瘤细胞凋亡的关系。方法采用免疫组化技术检测人多形性胶质母细胞瘤细胞系,经X射线处理前后p53、bcl-2基因表达的变化;原位杂交方法检测p53基因在mRNA水平表达的变化。结果①凋亡组内p53蛋白表达率明显高于对照组,bcl-2表达显著减弱,p53与bcl-2蛋白表达呈显著性负相关。②凋亡组内p53在mRNA水平表达量明显高于对照组,p53基因在蛋白水平与mRNA水平表达呈显著性正相关。结论p53和bcl-2基因在X射线诱导的细胞凋亡过程中具有重要作用
Objective To investigate the relationship between the changes of oncogene expression and apoptosis of tumor cells induced by radiation. Methods Immunohistochemistry was used to detect the expression of p53 and bcl-2 gene in human glioblastoma multiforme cell line before and after X-ray treatment. The expression of p53 gene at mRNA level was detected by in situ hybridization. Results ① The expression of p53 protein in the apoptosis group was significantly higher than that in the control group, the expression of bcl-2 was significantly weakened, and the expression of p53 and bcl-2 protein was negatively correlated. ② The expression level of p53 in the apoptosis group was significantly higher than that in the control group, p53 protein was significantly and positively correlated with the mRNA level. Conclusion The p53 and bcl-2 genes play an important role in the process of X-ray-induced apoptosis
