目的:探讨3种生长因子——干细胞因子(SCF)、白血病抑制因子(LIF)和碱性成纤维细胞生长因子(bF-GF)对精原干细胞(SSCs)体外存活及增殖的作用。方法:在培养液中加入不同浓度的SCF、LIF和bFGF,各生长因子均设浓度为0、5、10、20μg/L4个浓度组,每种生长因子不同浓度1式3份,每隔8~12h观察细胞的体外存活及增殖情况。应用光镜、电镜观察SSCs的形态结构特点。结果:培养液中加入不同浓度的SCF、LIF及bFGF后,SSCs的存活时间均比对照组有所延长,组间比较差异有显著性(P<0.05)。结论:生长因子SCF、LIF和bF-GF等可促进SSCs体外存活及增殖。
Objective: To investigate the effects of three growth factors - stem cell factor (SCF), leukemia inhibitory factor (LIF) and basic fibroblast growth factor (bF-GF) on the survival and proliferation of spermatogonial stem cells (SSCs) in vitro. Methods: Different concentrations of SCF, LIF and bFGF were added to the culture medium. Each growth factor was divided into 4 concentrations of 0, 5, 10 and 20 μg / L, and each concentration of growth factor was 1, 3, ~ 12h observation of cell survival and proliferation in vitro. Light microscopy and electron microscopy were used to observe the morphological characteristics of SSCs. Results: After the SCF, LIF and bFGF were added into the culture medium, the survival time of SSCs was longer than that of the control group. There was significant difference between the two groups (P <0.05). Conclusion: The growth factors SCF, LIF and bF-GF can promote the survival and proliferation of SSCs in vitro.
