采用荧光染色、双抗体标记染色和流式细胞分析技术,对55例颌面部鳞癌、乳头状瘤及正常面部皮肤组织进行DNA指数、增殖指数、S期细胞比例及p53基因产物表达的定量分析。结果:正常皮肤和乳头状瘤均为二倍体,而鳞癌异倍体率为60%,增殖指数和S期细胞所占的比例在这3种组织中均有明显的差异;在正常组织无p53阳性表达,乳头状瘤p53的阳性表达率在30%左右,鳞癌的阳性表达率为62.5%;鳞癌p53的阳性表达率随病理组织学分级的增高而增高,但与肿瘤细胞生物学特性无相关性。根据结果可以认为p53基因产物的表达在良恶性肿瘤组织中有非常显著性差异,并与肿瘤的恶性度有一定关系,因此,可将p53蛋白的检测作为良恶性肿瘤鉴别的辅助指标。
Fifty-five patients with squamous cell carcinoma, papilloma and normal facial tissues of the maxillofacial region were screened for DNA index, proliferation index, proportion of S phase cells and expression of p53 gene product using fluorescent staining, double antibody labeling and flow cytometry analysis. Results: The normal skin and papilloma were diploid, while the rate of aneuploidy in squamous cell carcinoma was 60%. The proliferation index and the proportion of S phase cells were significantly different among the three tissues. In normal tissues The positive rate of p53 in papilloma was about 30% and the positive rate in squamous cell carcinoma was 62.5%. The positive rate of p53 in squamous cell carcinoma increased with the increase of pathological grade, No correlation was found between cell biological characteristics. According to the results, it can be considered that the expression of p53 gene product has a very significant difference in benign and malignant tumor tissues and has a certain relationship with the malignancy of the tumor. Therefore, the detection of p53 protein can be used as an auxiliary indicator for the identification of benign and malignant tumors.
