目的研究千年健超临界CO2萃取及萃取物包合的最佳工艺,并测定萃取物的化学成分。方法采用均匀设计方法,以萃取压力、萃取温度、萃取时间为因素,考察超临界CO2萃取的最佳工艺条件并对萃取物进行GC-MS分析;采用正交设计方法,以包合物收率,包合物油利用率、包合物含油率为指标,研究主客分子比、碾磨时间、加水倍量、加醇量对包合结果的影响并进行药效学研究。结果超临界CO2萃取工艺为:压力20MPa,温度60℃,时间30min;包合最佳工艺为主客分子比1∶4,研磨30min,加入2倍量的水,含醇量20%;萃取物包合物具有显著的抗炎、镇痛作用;所得萃取物主要成分为芳樟醇。结论利用超临界CO2萃取技术萃取千年健,可以缩短萃取时间,明显提高收率;优选出的包合工艺参数稳定,可行。
Objective To study the optimum conditions for the extraction of millennium health supercritical carbon dioxide (CO2) and the inclusion of the extract, and to determine the chemical composition of the extract. Methods The uniform design method was used to investigate the optimum extraction conditions of supercritical carbon dioxide (CO2) extraction by extraction pressure, extraction temperature and extraction time. GC-MS analysis of the extracts was carried out. The orthogonal design was used to determine the inclusion compound yield , Inclusion oil utilization and oil content of inclusion compound were used as indices to study the influence of host-guest molecular ratio, milling time, water addition volume and added alcohol content on the inclusion results and to conduct pharmacodynamic studies. Results The supercritical CO2 extraction process was as follows: the pressure was 20MPa, the temperature was 60 ℃, the time was 30min. The optimal process of inclusion was the host-guest ratio of 1: 4, grinding for 30min, adding 2 times of water, The compound has significant anti-inflammatory and analgesic effects. The main component of the obtained extract is linalool. Conclusion The extraction of millennium health by supercritical CO2 extraction can shorten the extraction time and improve the yield obviously. The optimal inclusion process parameters are stable and feasible.
