采用超临界CO_2萃取法提取猴头菌中油脂类成分,以提取率为评价指标,提取压力、温度和挟带剂量为变量,优化超临界提取工艺。猴头油脂类成分甲酯化后用气相色谱-质谱联用技术分析脂肪酸组成,并以37种脂肪酸混合标准品为参照物,外标法进行定量分析。结果表明,超临界的提取压力和挟带剂量为提取率的显著影响因素,最优提取条件为压力40 MPa、温度35℃、挟带剂量20 mL,得率为7.59%。猴头菌超临界CO_2萃取法提取物中最多检测出14种脂肪酸,其中不饱和脂肪酸有4种,以亚油酸和油酸的含量较高。组合8、9和追加组的提取率较高,其所含的脂肪酸量基本相近。组合9含量略高,其不饱和脂肪酸含量约占总脂肪酸的83%,其中多不饱和脂肪酸亚油酸含量最高,达到281.98μg/g,单不饱和脂肪酸油酸次之,含量为193.25μg/g。该研究为猴头菌的质量评价及其保健食品研发提供实验依据。
The supercritical CO_2 extraction of Hericium oil components, the extraction rate as an evaluation index, extraction pressure, temperature and entrained dose as variables, optimizing the supercritical extraction process. Herbicidal composition was esterified and analyzed by gas chromatography-mass spectrometry (GC-MS). The fatty acid composition was analyzed by 37 fatty acid mixed standards and the external standard method was used for quantitative analysis. The results showed that the supercritical extraction pressure and entrained dose were significant influencing factors of extraction rate. The optimal extraction conditions were pressure 40 MPa, temperature 35 ℃, entrained dose 20 mL, yield 7.59%. Hericium erinaceus supercritical CO_2 extract detected up to 14 kinds of fatty acids, including four kinds of unsaturated fatty acids, linoleic acid and oleic acid content is higher. The combination of 8,9 and the additional group higher extraction rate, which contains the amount of fatty acids are basically similar. The contents of unsaturated fatty acids were about 83% of the total fatty acids, the content of linoleic acid was the highest, reaching 281.98μg / g, followed by monounsaturated fatty acid oleic acid, the content was 193.25μg / g. The research provided the experimental evidence for the quality evaluation of Hericium erinaceus and the research and development of its health food.
