目的:观察PP1脂质体滴眼对大鼠眼的急性毒性作用。方法:用10,60,400μmol·L~(-1)低、中、高3个不同浓度的PP1脂质体滴入SD大鼠单侧眼,对侧眼给予空载脂质体作为对照,4次/d。在裂隙灯显微镜下观察眼结膜、角膜和虹膜的改变,连续观察3d,用眼前节刺激评分表评分。1周后取大鼠眼角膜、虹膜和晶状体组织,分别在光镜下观察各组织的病理变化,在电镜下观察角膜上皮细胞与内皮细胞的超微结构变化。结果:在浓度为400μmol·L~(-1)的PP1脂质体滴眼后第1天,1只大鼠出现少许结膜分泌物,其余各组大鼠均未出现刺激反应,各组大鼠角膜荧光素钠染色均为阴性;不同浓度PP1脂质体滴眼后大鼠的角膜、虹膜、晶状体组织结构无异常改变,角膜上皮细胞与内皮细胞超微结构亦无异常改变。结论:低、中、高3个不同浓度的PP1脂质体点眼对SD大鼠无急性眼毒性作用。
Objective: To observe the acute toxic effects of PP1 liposome eye drops on rat eyes. Methods: The low-dose, middle-high and high-concentration PP1 liposomes were injected into the unilateral eyes of SD rats at 10, 60 and 400μmol·L -1, respectively. Times / d. Observed in the slit lamp microscope conjunctiva, corneal and iris changes, continuous observation 3d, with the front section of the stimulus score score. After 1 week, the cornea, iris and lens were taken from the cornea, the pathological changes of each tissue were observed under light microscope, and the ultrastructural changes of corneal epithelial cells and endothelial cells were observed under electron microscope. Results: On the first day after PP1 liposome instillation with a concentration of 400 μmol·L -1, a little conjunctival secretions were observed in one rat and no irritation was observed in the other rats. Rats in each group Corneal fluorescein sodium staining were negative; different concentrations of PP1 liposome eye drops rat cornea, iris, lens structure of the abnormal changes in the corneal epithelial cells and endothelial cell ultrastructure did not change abnormally. Conclusion: There are no acute ocular toxicity effects of PP1 liposomes with low, medium and high concentrations on the SD rats.
