目的:建立高效液相色谱法测定氯达软膏中氯霉素的含量。方法:色谱柱为Hypersil ODS C_(18)柱(150 mm×4.6mm,5μm),流动相为含0.1%庚烷磺酸钠的溶液(取0.1%庚烷磺酸钠溶液500ml与二甲基甲酰胺5ml、冰醋酸0.5ml,混匀)-乙腈(75:25),流速为1.0ml·min~(-1),检测波长为272nm,进样量为20μl。结果:氯霉素浓度在25.6~128.4μg·ml~(-1)范围内与峰面积具有良好线性关系,r=0.999 9,平均回收率为99.7%(RSD=0.9%,n=9)。结论:本法简便、结果准确、重复性好,可用于氯达软膏的的质量控制。
Objective: To establish a HPLC method for the determination of chloramphenicol in Chlordecone ointment. METHODS: The chromatographic column was Hypersil ODS C 18 column (150 mm × 4.6 mm, 5 μm). The mobile phase consisted of 0.1% sodium heptanesulfonate solution (500 ml of 0.1% sodium heptanesulfonate and dimethyl Formamide 5ml, glacial acetic acid 0.5ml, mixed) -acetonitrile (75:25) at a flow rate of 1.0ml · min -1. The detection wavelength was 272nm and the injection volume was 20μl. Results: The linear range of chloramphenicol to peak area was 25.6 ~ 128.4μg · ml ~ (-1), r = 0.999 9, the average recovery was 99.7% (RSD = 0.9%, n = 9). Conclusion: This method is simple, accurate and reproducible. It can be used for the quality control of chlorine ointment.
