目的:观察补肺健脾方对慢性阻塞性肺疾病(COPD)大鼠骨骼肌Fas/FasL/Caspase-3的影响。方法:SD大鼠48只随机分为对照组、模型组、补肺健脾组和氨茶碱组,每组12只。采用香烟熏吸联合细菌反复感染法制备COPD稳定期大鼠模型。自第9周起,对照组和模型组均给予0.9%氯化钠溶液灌胃,其余2组分别给予补肺健脾方及氨茶碱治疗,第20周结束时取材。采用TUNEL技术检测股四头肌、肋间肌、肱二头肌和比目鱼肌4种骨骼肌的细胞凋亡率;采用荧光定量PCR技术和Western blot技术观察4种骨骼肌Fas、FasL和Caspase-3 mRNA和蛋白表达。结果:模型组4种肌肉细胞凋亡率和Fas、FasL、Caspase-3表达均较对照组显著升高(P<0.01),补肺健脾方和氨茶碱均可显著改善上述指标(P<0.05,P<0.01)。结论:补肺健脾方可以显著改善COPD大鼠骨骼肌凋亡,降低Fas、FasL和Caspase-3 mRNA和蛋白表达。
Objective: To observe the effects of Bufei Jianpi Recipe on skeletal muscle Fas / FasL / Caspase-3 in rats with chronic obstructive pulmonary disease (COPD). Methods: Forty-eight SD rats were randomly divided into control group, model group, Bufei Jianpi group and aminophylline group, with 12 rats in each group. COPD stable rat model was prepared by cigarette smoking combined with bacterial repeated infection. From the 9th week onward, 0.9% sodium chloride solution was given to the control group and model group. The other two groups were treated with Bufei Jianpi Fang and aminophylline, respectively. TUNEL technique was used to detect the apoptosis rate of four kinds of skeletal muscle of quadriceps, intercostal muscles, biceps and soleus muscle. The expressions of Fas, FasL and Caspase- 3 mRNA and protein expression. Results: The apoptosis rate of four kinds of muscle cells and the expressions of Fas, FasL and Caspase-3 in the model group were significantly higher than those in the control group (P <0.01). Both Bufei Jianpi Fang and aminophylline significantly improved the above indexes <0.05, P <0.01). Conclusion: Bufei Jianpi Fang can significantly improve the apoptosis of skeletal muscle of COPD rats and decrease the expression of Fas, FasL and Caspase-3 mRNA and protein.
