目的研究贵州地区汉族人群HPA-1~5、15基因多态性分布特点,为本地区HPA免疫性疾病的防治和HPA基因分型库的建立提供参考数据。方法采用PCR-SSP方法对本地区700例汉族人群无血缘关系的健康献血员进行HPA-1~5、15系统基因分型,计算基因型及基因频率,总结出多态性分布特点,并与国内其他地区和国外不同人种进行比较,阐明本地区特点。结果各系统均呈多态性分布。HPA-1和-4接近aa基因型单线分布;HPA-2和-5少量杂合,ab杂合子依次为64和30例,4例-2bb;HPA-3和-15高度杂合,aa基因型依次为242和210例,ab型依次为347和350例,且有较多的bb基因型,依次为111和140例。与其他地区和不同国家人群人种的比较均或多或少存在差异,并随地域和人种的差别而增加。结论贵州地区汉族人群HPA-1~5、15系统基因均呈多态性分布,以HPA-15、-3、-2和-5的a与b基因杂合率较高,对于临床HPA免疫性疾病的防治应给予高度重视。
Objective To investigate the distribution of HPA-1 ~ 5,15 gene polymorphism in Guizhou Han population and to provide reference data for the prevention and treatment of HPA immunodeficiency disease and the establishment of HPA genotyping library in this area. Methods PCR-SSP method was used to genotype HPA-1 ~ 5 and 15 healthy unrelated blood donors in 700 Han nationality in this area. The genotype and gene frequency were calculated and the polymorphism distribution was summarized. Other regions and different ethnic groups to compare, explain the characteristics of the region. Results All the systems showed polymorphism distribution. HPA-1 and -4 were close to the aa genotype. HPA-2 and -5 were minor heterozygotes. The number of heterozygotes for abA was 64 and 30, and the number of HPA-3 and-15 was 4 and 2b respectively. Followed by 242 and 210 cases, ab type followed by 347 and 350 cases, and there are more bb genotypes, followed by 111 and 140 cases. Compared with the ethnic groups in other regions and different countries are more or less different, and with the geographical and ethnic differences. CONCLUSIONS: The HPA-1 ~ 5,15 genes in Han population of Guizhou are polymorphic. The heterozygosity of a and b genes of HPA-15, -3, -2 and -5 is high, and the clinical HPA immunity Disease prevention and treatment should be given high priority.
