目的观察浙江主产区栽培玄参的群体遗传结构。方法应用荧光AFLP标记对具有代表性的6个玄参居群群体遗传结构进行分析。结果采用7对AFLP引物组合扩增共得到12 552条扩增带,其中多态性谱带8 808条,多态性比率70.17%;各居群间的PPB变化范围在41.67%~55.56%,平均为47.30%;I在0.190 8~0.238 3,平均为0.221 8;Ne在1.201 4~1.280 6,平均为1.236 9;Gst为0.127 1,Nm为3.432 4;UPGMA聚类显示6个居群可以聚为2类,其中天台、缙云和景宁种群遗传距离较近聚为一个亚类,东阳、磐安和仙居聚为一个亚类。结论浙江主产区栽培玄参种质资源在物种水平上具有较高的遗传多样性,居群间有一定的遗传分化,但大部分存在于居群内部,居群间有较高的基因交流,居群间遗传距离与地理环境无相关性。
Objective To observe the population genetic structure of cultivated Scrophulariaceae in the main producing areas of Zhejiang Province. Methods Fluorescent AFLP markers were used to analyze the genetic structure of six representative populations of Scrophularia ningpoensis population. Results A total of 12 552 bands were amplified using 7 AFLP primer combinations, of which 8 808 were polymorphic bands with a polymorphism rate of 70.17%. Conclusion The genus Scrophulariaceae cultivars cultivated in the main producing areas of Zhejiang Province have high genetic diversity at the species level, with some genetic differentiation among the populations, but most of them exist within the populations and have high gene communication among the populations There was no correlation between genetic distance and geographical environment.
