目的:研究青阳参葡聚糖硫酸酯体外抗艾滋病毒活性及作用机制。方法:以合胞体形成抑制试验和HIV-1 p24抗原ELISA检测法,测定其抗HIV-1的活性;以时间过程试验、细胞融合试验和预处理试验等探寻作用机制。结果:青阳参葡聚糖硫酸酯抗HIV-1ⅢB,HIV-1Ada-M和HIV-1Bal的半数抑制浓度(IC50)分别为0.26,0.46,0.90μmol.L-1;作用机制研究显示,青阳参葡聚糖硫酸酯作用于HIV感染细胞的早期阶段。结论:青阳参葡聚糖硫酸酯具有较高的抗HIV-1活性,可能是通过与病毒的囊膜蛋白的相互作用而发挥抗病毒活性。
Objective: To study the anti-HIV activity and mechanism of Qingyang ginseng dextran sulfate in vitro. Methods: The anti-HIV-1 activity was determined by inhibition of syncytium formation and HIV-1 p24 antigen ELISA. The mechanism of action was investigated by time course test, cell fusion test and pretreatment test. Results: The median inhibitory concentration (IC50) of anti-HIV-1ⅢB, HIV-1Ada-M and HIV-1Bal were 0.26,0.46 and 0.90μmol.L-1, respectively. The sun-ginseng dextran sulfate acts on the early stages of HIV-infected cells. CONCLUSIONS: Dextran sulfate has high anti-HIV-1 activity, which may exert its antiviral activity through the interaction with the viral envelope protein.
