目的探讨高脂饮食对兔肺动脉组织形态学、平滑肌增殖和肺小动脉肌化的影响。方法建立高脂兔模型,对肺动脉及肺组织石蜡切片进行α-smoothmuscle-actin、增殖细胞核抗原(PCNA)免疫组化检查及HE染色结合形态计量分析观察肺血管重构。结果高脂饮食组肺动脉粥样硬化面积占血管内膜面积的比值、中膜厚度、小动脉肌化比例、中膜平滑肌细胞增殖指数均明显高于对照组。结论高脂饮食时除了肺动脉出现粥样硬化外,肺动脉平滑肌细胞增殖增加、肺血管出现重构。
Objective To investigate the effects of high-fat diet on pulmonary arterial morphology, smooth muscle proliferation and pulmonary arterioles in rabbits. Methods The model of hyperlipidemic rabbits was established. Paraffin sections of pulmonary artery and lung tissue were examined with α-smooth muscle actin, proliferating cell nuclear antigen (PCNA) immunohistochemistry and HE staining combined with morphometric analysis to observe pulmonary vascular remodeling. Results The ratio of pulmonary atherosclerosis area to intima - media thickness, medial thickness, arteriolar arterialization ratio, and smooth muscle cell proliferation index in the high - fat diet group were significantly higher than those in the control group. Conclusion In addition to pulmonary atherosclerosis in high-fat diet, pulmonary artery smooth muscle cells increased proliferation, pulmonary vascular remodeling.
