背景:有多种因素对大鼠前列腺增生模型影响的实验研究,但抑郁对大鼠前列腺增生的影响报道较少。目的:探讨抑郁因素对大鼠前列腺增生的影响。设计:随机对照的实验研究。地点和材料:实验地点:首都医科大学宣武医院动物室;雄性SD(Sprague-Dawleg)大鼠40只,体质量220~230g。为SPF级无特定病原体动物。干预:将大鼠分为正常对照组、抑郁组、丙酸睾丸酮组及抑郁+丙酸睾丸酮组,抑郁组及抑郁+丙酸睾丸酮组在大鼠颈部套一红色圆形项夹,丙酸睾丸酮组及抑郁+丙酸睾丸酮组按1mg/(只·d)皮下注射丙酸睾丸酮(丙酸睾丸酮溶于大豆油,2g/L),正常对照组和抑郁组注射等量溶媒,造模3周。主要观察指标:测定大鼠前列腺质量,前列腺指数,行苏木精-伊红染色观察形态学改变,组织增殖细胞核抗原(proliferatingcellnuclearanti-gen,PCNA)免疫组化染色计算PCNA增殖指数。结果:抑郁组,丙酸睾丸酮组及抑郁+丙酸睾丸酮组前列腺质量、前列腺指数均显著高于正常对照组(P<0.05~0.01),丙酸睾丸酮组及抑郁+丙酸睾丸酮组前列腺质量及前列腺指数显著高于单纯抑郁组(P<0.01)。单纯抑郁组前列腺指数及PCNA增殖指数显著高于对照组(P<0.05),组织学观察抑郁组前列腺有增生的表现。结论:抑郁对前列腺增生有一定的诱发作用。
BACKGROUND: There are many factors that influence the experimental model of benign prostatic hyperplasia in rats, but there are few reports of the effects of depression on prostatic hyperplasia in rats. Objective: To investigate the effect of depression on the prostatic hyperplasia in rats. Design: Randomized controlled experimental study. Venue and material: Experimental sites: Xuanwu Hospital, Capital Medical University animal room; male SD (Sprague-Dawleg) 40 rats, body weight 220 ~ 230g. For the SPF level no specific pathogen animals. Intervention: The rats were divided into normal control group, depression group, testosterone propionate group, depression + testosterone propionate group, depression group and depression + testosterone propionate group in the neck of rats with a red circular clip, propionic acid Testosterone propionate group and testosterone propionate group were subcutaneously injected with testosterone propionate (testosterone propionate dissolved in soybean oil, 2g / L) at 1mg / (d · d), and normal control group and depression group were injected with equal amount of vehicle week. MAIN OUTCOME MEASURES: Prostate mass, prostate index, morphological changes of hematoxylin-eosin staining and proliferating cell nuclear antigen (PCNA) immunohistochemistry were used to determine PCNA proliferation index. Results: Prostate mass and prostate index in depression group, testosterone propionate group and depression + testosterone propionate group were significantly higher than those in normal control group (P <0.05-0.01), prostate testosterone propionate group and depression + testosterone propionate group Prostate index was significantly higher than that of simple depression group (P <0.01). Prostate index and proliferative index of PCNA in simple depression group were significantly higher than those in control group (P <0.05). Histological observation showed that prostate hyperplasia in depression group. Conclusion: Depression has a certain inducing effect on prostatic hyperplasia.
