【目的】甘薯对人体有较强的保健作用,特别是富含胡萝卜素的品种保健价值更高。采用分子标记和农艺性状相结合评价亲本亲缘关系,可提高甘薯高胡萝卜素食用品种的选育效果。【方法】评价15个亲本及其集团杂交后代的农艺性状、品质性状,结合RAPD、ISSR分子标记,进行聚类分析,筛选亲本。【结果】亲本间胡萝卜素含量和鲜薯产量两个重要性状的变异范围较大。胡萝卜素含量与鲜薯产量和薯干产量均呈负相关。可溶性糖、蛋白质、胡萝卜素含量几个重要的品质性状之间未发现不利相关现象。亲子代间胡萝卜素含量呈极显著正相关,相关系数为0.7932**。基于农艺性状分析供试材料亲缘关系与已知系谱不吻合,基于分子标记聚类分析结果与已知系谱吻合度较高;ISSR标记每条引物平均扩增条带13.8条,多态性条带比率为89.86%,RAPD标记每条引物平均扩增条带9.4条,多态性条带比率为74.46%。【结论】甘薯常用亲本胡萝卜素含量遗传背景丰富;利用分子标记分析亲本亲缘关系比农艺性状客观;ISSR标记比RAPD标记分析效率高;结合分子标记和农艺性状,可为甘薯高胡萝卜素食用品种选育亲本组配提供一定的理论依据。
【Objective】 Sweet potato has a strong health effect on the human body, especially the carotene-rich varieties have higher health value. The combination of molecular markers and agronomic traits to evaluate the genetic relationship of parents can improve the breeding effect of sweetpotato high-carotene variety. 【Method】 The agronomic traits and quality traits of 15 parents and their group hybrids were evaluated. RAPD and ISSR markers were used for cluster analysis and the parents were screened. 【Result】 The range of variation of two important traits, carotene content and fresh potato yield, was large. Carotene content and fresh potato production and dry production were negative correlation. Soluble sugar, protein, carotene content of several important quality traits found no adverse correlation between the phenomenon. There was a significant and positive correlation between carotene content between parents and offspring, the correlation coefficient was 0.7932 **. Based on the analysis of agronomic traits, the genetic relationship of tested materials was not consistent with the known pedigree. The results of cluster analysis based on molecular markers showed high agreement with the known pedigree. The average number of amplified bands per primer was 13.8, and the polymorphic bands The rate was 89.86%. The average number of bands amplified by RAPD markers was 9.4 and the percentage of polymorphic bands was 74.46%. 【Conclusion】 The common parental carotene content in sweet potato is rich in genetic background. The genetic relationship of parents is better than that of agronomic traits by molecular markers. ISSR markers are more efficient than RAPD markers. Combined with molecular markers and agronomic traits, Parenting group to provide some theoretical basis.
