目的 研究10 3 Pd粒子与加热 41℃对人肝癌细胞株 (Bel -740 2 )的细胞毒作用 ,了解10 3 Pd粒子对热疗后残存的肝癌细胞的细胞毒作用。方法 以体外培养的人肝癌细胞 -740 2为研究对象 ,采用对照分组、水浴加温法、体外细胞毒试验 (MTT)、凋亡实验 (Hoechst3 3 2 5 8荧光染色法 )的方法以及方差分析 ,观察10 3 Pd粒子放疗联合加热 41℃对Bel -740 2细胞的生长抑制作用。结果 10 3Pd粒子对 41℃加热后的人肝癌细胞 -740 2有明显的细胞抑制作用 ,强于单纯10 3 Pd粒子放疗。结论 人肝癌细胞 -740 2对10 3 Pd粒子放疗敏感 ,10 3 Pd粒子联合加热 41℃有增效作用。提示 :10 3 Pd粒子对热疗后残存的肝癌细胞有细胞毒作用。
Objective To study the cytotoxic effect of 10 3 Pd particles and heating at 41 ℃ on human hepatoma cell line Bel -740 2 and to understand the cytotoxic effect of 10 3 Pd particles on remaining hepatoma cells after hyperthermia. Methods Human hepatocellular carcinoma cell line-7402 was cultured in vitro. The cells were divided into two groups: control group, water bath warming method, MTT assay, Hoechst 33258 fluorescence staining method and analysis of variance The growth inhibition of Bel-7402 cells by 10 3 Pd particle radiotherapy combined with heating at 41 ℃ was observed. Results 10 3Pd particles had a significant cytostatic effect on human hepatoma cell-7402 after 41 ℃ heating, which was stronger than that of pure 10 3 Pd particle radiotherapy. Conclusion Human hepatoma cell-7402 is sensitive to 10 3 Pd particles radiotherapy, and 10 3 Pd particles combined with heating at 41 ℃ have synergistic effects. Tip: 10 3 Pd particles on the residual liver cancer cells after cytotoxic effect.
