目的:建立金银花指纹图谱混合酸流动相测定方法。方法:采用高效液相色谱法,色谱柱为DIKMA Diamonsil C18(2)色谱柱(4.6 mm×250 mm,5μm),以乙腈为流动相A,以含0.025%磷酸、0.3%醋酸的溶液为流动相B,进行梯度洗脱;检测波长为243 nm。柱温为40℃。结果:使用混合酸能改善30~50 min各色谱峰的分离度,使52 min混合峰完全基线分离。结论:混合酸流动相使金银花色谱峰分离度增加,可用于金银花指纹图谱的研究。
Objective: To establish a method for the determination of the mixed acid mobile phase of the honeysuckle fingerprints. Methods: High performance liquid chromatography (HPLC) was performed on a DIKMA Diamonsil C18 (2) column (4.6 mm × 250 mm, 5 μm) using acetonitrile as mobile phase A and a solution containing 0.025% phosphoric acid and 0.3% acetic acid Phase B, gradient elution; detection wavelength of 243 nm. The column temperature was 40 ° C. Results: The mixed acid could improve the resolution of each chromatographic peak from 30 to 50 min, and completely separate the 52 min mixed peak from baseline. Conclusion: Mixed acid mobile phase can increase the resolution of Honeysuckle chromatographic peaks, which can be used for the study of fingerprints of Honeysuckle.
