目的 研究短波紫外线照射对恶性肿瘤患者粒 -单造血祖细胞集落形成和细胞活力的影响。方法 应用 0 ,5 0 ,10 0 ,30 0 ,40 0 ,80 0mJ/cm2 的UVC照射应用干细胞生长因子动员后白细胞外周血单个核细胞 (PBMNC) ,观测 7d后粒 -单系造血祖细胞集落 (CFU -GM)以及于UVC照射后1,3,5 ,7d观察细胞活力。结果 5 0 ,10 0 ,2 0 0 ,40 0 ,80 0mJ/cm2 UVC照射后CFU -GM集落个数分别平均为 37.7,16 .0 ,7.4,4.7,0 .8,与对照组 (5 9.5 )比较 ,集落形成率差异有非常显著性 (χ2 =5 2 .0 5 ,P =0 .0 1)。于照射后 1,3,5 ,7d观察细胞成活率 ,在UVC照射后 1d和 3d细胞活力的下降与对照组比较差异无显著性 (P >0 .0 5 ) ,第 5d和第 7d 5 0mJ/cm2 UVC照射与对照组比较差异无显著性 ,而 >10 0mJ/cm2 的降低与对照组差异有非常显著性 (P <0 .0 1)。结论 UVC照射可显著影响粒 -单系造血祖细胞集落形成 ,照射对细胞的影响并非直接对细胞的杀伤 ,可能是通过改变细胞膜受体而影响细胞的功能。
Objective To study the effect of shortwave UV irradiation on the colony formation and cell viability of granulocyte-mononuclear progenitor cells in patients with malignant tumors. Methods Peripheral blood mononuclear cells (PBMNC) were mobilized by UVC of 0, 5 0, 10 0, 30 0, 40 0 and 80 0 mJ / cm2. After 7 days, the granulocyte-mononuclear hematopoietic progenitor cells (CFU -GM) and 1, 3, 5, 7 days after UVC irradiation. Conclusion UVC irradiation can significantly affect the granulocyte - monocyte lineage hematopoietic progenitor colony formation, irradiation on the cells is not directly on the cell killing, may affect the cell function by changing the cell membrane receptors.
