目的:研究麦门冬汤对人肺腺癌A549细胞凋亡的影响。方法:采用噻唑蓝(MTT)法检测麦门冬汤(0.25、2.5、25g/L)作用24、48、72h后人肺腺癌A549细胞(2×104个/m L)活性;吖啶橙(AO)/EB双荧光染色观察麦门冬汤各剂量作用48h后A549细胞凋亡形态学改变;流式细胞术分析麦门冬汤作用48h后A549细胞凋亡率;Western Blot检测麦门冬汤对凋亡相关蛋白表皮生长因子受体(EGFR)、信号转导子和转录激活子3(STAT3)表达的影响。结果:各剂量麦门冬汤能够抑制A549细胞活性,诱导其发生凋亡形态学改变,且A549细胞凋亡率与麦门冬汤剂量呈正相关(P<0.01);Western Blot检测表明低、中剂量麦门冬汤可使EGFR、STAT3表达下降。结论:麦门冬汤可诱导A549细胞凋亡,其作用机制可能与下调EGFR、STAT3蛋白表达相关。
Objective: To study the effect of Mai Men Dong decoction on the apoptosis of human lung adenocarcinoma A549 cells. Methods: The activity of human lung adenocarcinoma A549 cells (2 × 104 cells / m L) after 24, 48, and 72 hours were treated with 0.25, 2.5, 25 g / L of Mai Mintong Decoction (MTT) (AO) / EB double staining was used to observe the morphological changes of apoptosis in A549 cells treated with different dosages of Mai Mendong Decoction for 48 h; the apoptosis rate of A549 cells was analyzed by flow cytometry after 48 h; Effects of Decoction on Expressions of Epidermal Growth Factor Receptor (EGFR), Signal Transducer and Activator of Transcription Activator 3 (STAT3) in Apoptosis Related Protein. Results: All doses of Mai Menteng Decoction could inhibit the activity of A549 cells and induce morphological changes of apoptosis, and the apoptosis rate of A549 cells was positively correlated with the dose of Mai Men Dong Decoction (P <0.01) Dose Mai Mendong Decoction can make EGFR, STAT3 expression decreased. Conclusion: Mai-meng-dong decoction can induce the apoptosis of A549 cells, which may be related to the down-regulation of EGFR and STAT3 protein expression.
