目的建立测定头孢克肟含量的胶束电动毛细管色谱方法。方法采用非涂渍石英毛细管51cm×75μm;运行缓冲液为含100mmol·L-1十二烷基硫酸钠的磷酸盐缓冲液(70mmol·L-1,pH7.0);压力进样5s,分离电压15kV,柱温25℃,检测波长254nm,选择乙酰苯胺为内标。结果头孢克肟在6.08~777.98μg·mL-1内,线性关系良好(r=0.9993)。头孢克肟的最低检测限为1.03μg·mL-1,最低定量限为3.43μg·mL-1;含量测定结果与HPLC所得结果没有显著性差异。结论胶束电动毛细管色谱法分析头孢克肟的含量,方法准确、简便、灵敏,与HPLC具有互补性。
Objective To establish a method for the determination of cefixime by micellar electrokinetic capillary chromatography. Methods Non-coated quartz capillary 51cm × 75μm was used. Running buffer was phosphate buffer (70mmol·L-1, pH7.0) containing 100mmol·L-1 sodium lauryl sulfate. Voltage 15kV, column temperature 25 ℃, detection wavelength 254nm, select acetanilide as internal standard. Results The calibration curve of cefixime in 6.08 ~ 777.98μg · mL-1 was good (r = 0.9993). The detection limit of cefixime was 1.03μg · mL-1, the lowest limit of quantification was 3.43μg · mL-1. There was no significant difference between the results of determination and HPLC. Conclusion Micellar electrokinetic capillary chromatography analysis of cefixime content, accurate, simple, sensitive, and HPLC are complementary.
