目的:探讨细粒棘球蚴(Echinococcus granulosus,Eg)囊液(HCF)对大鼠肝星状细胞(HSC)T6细胞系增殖的影响。方法:HCF体外刺激T6细胞,通过MTT法检测细胞增殖,流式细胞仪检测细胞周期,Western Blot检测细胞外调节激酶(ERK1/2)磷酸化、细胞增殖核抗原(PCNA)及细胞周期蛋白(cyclin)D1、E、A和B1表达水平。结果:6 h和12 h,15%、30%和60%HCF组与对照组相比增殖显著(P<0.05);15%HCF组细胞增殖指数分别为57.9%和71%;Western Blot检测显示15%HCF组pERK、PCNA和cyclin E在0.5 h时的表达量较为对照组差异显著(P<0.05);cyclin D1在6 h时表达量较对照组差异显著(P<0.05)。结论:HCF可能是通过激活ERK后调控细胞周期相关蛋白的表达促进了HSC的增殖。
Objective: To investigate the effects of Echinococcus granulosus (Eg) cystic fluid (HCF) on the proliferation of rat hepatic stellate cells (HSC) T6 cell line. Methods: HCT stimulated T6 cells in vitro, the proliferation of T6 cells was detected by MTT assay, the cell cycle was detected by flow cytometry, the phosphorylation of ERK1 / 2, the expression of proliferating cell nuclear antigen (PCNA) and cyclin cyclin) D1, E, A and B1 expression levels. Results: The proliferation of HCF group was significantly higher than that of the control group at 6 h, 12 h, 15%, 30% and 60% (P <0.05). The proliferation index of 15% HCF group was 57.9% and 71% The expression of pERK, PCNA and cyclin E in 15% HCF group was significantly different from that in control group at 0.5 h (P <0.05). The expression of cyclin D1 at 6 h was significantly different from that in control group (P <0.05). Conclusion: HCF may promote the proliferation of HSC by regulating the expression of cell cycle related proteins after activating ERK.
