小鼠胸苷激酶缺陷型细胞(LTK~-)在遗传工程,在生物化学及病毒学领域有广泛的用途。本实验对(LTK~-)细胞的核型进行了分析(C、G、Ag显带),结果表明LTK~-细胞染色体的数量与形态都与正常小鼠细胞有很大差别。LTK-细胞的染色体众数为49,其中有12~16条为中或亚中着丝粒染色体;银染颗粒众数为6对。与正常小鼠染色体比较,染色体的长度、着丝粒位置、数目等均有显著差异,可能是由亍倒位,易位、丢失等染色体结构变化引起的、本实验说明LTK~-细胞是转化了的细胞系。
Mouse thymidine kinase-deficient cells (LTK ~ -) are widely used in genetic engineering and biochemistry and virology. In this experiment, the karyotypes of (LTK ~ -) cells were analyzed (C, G, Ag banding). The results showed that the number and morphology of LTK ~ - chromosomes were all different from those of normal mice. The chromosome number of LTK- cells was 49, of which 12-16 were middle or sub-centromeric chromosomes; the number of silver-stained particles was 6 pairs. Compared with the normal mouse chromosomes, there are significant differences in chromosome length, centromere location and number, which may be caused by changes in the chromosome structure such as inversion, translocation and loss.
