目的探讨核因子κB(NF-κB)诱捕脱氧寡核苷酸(ODNs)对子宫内膜息肉(EP)间质细胞在白细胞介素1β(IL-1β)诱导培养下环氧化酶2(COX-2)合成及雌激素(E2)分泌的影响。方法 15例EP患者行间质细胞的分离培养,分为3组:刺激(A)组,用10μg/L的IL-1β刺激培养;转染后刺激(B)组,NF-κB诱捕ODNs转染EP间质细胞后,用10μg/LIL-1β刺激培养;空白对照(C)组。培养12、24、36和48h,RT-PCR检测细胞中COX-2水平,ELISA检测细胞培养上清中E2水平。结果 A组自12h开始COX-2合成显著高于C组,自24h开始E2的分泌量显著高于C组(P<0.01)。B组COX-2合成及E2分泌较A组显著下降(P<0.01)。结论 NF-κB诱捕ODNs能显著抑制IL-1β诱导的EP间质细胞合成COX-2及分泌E2。
Objective To investigate the effect of NF-κB decoy oligodeoxynucleotides (ODNs) on the expression of cyclooxygenase-2 (COX) in endometrial polyps (EP) stromal cells under induction of IL- -2) synthesis and estrogen (E2) secretion. Methods Fifteen patients with EP were divided into three groups: group A stimulated with 10 μg / L IL-1β, group B stimulated with NF-κB After staining with EP mesenchymal cells, the cells were stimulated with 10μg / L IL-1β, and blank control group (C). Cultures were cultured for 12, 24, 36 and 48 hours. COX-2 levels in the cells were detected by RT-PCR and E2 in the cell culture supernatants by ELISA. Results The COX-2 synthesis in group A was significantly higher than that in group C at 12h. The secretion of E2 was significantly higher than that in group C at 24h (P <0.01). The COX-2 synthesis and E2 secretion in group B were significantly lower than those in group A (P <0.01). Conclusion ODNs can significantly inhibit COX-2 secretion and E2 secretion induced by IL-1β in EP stromal cells.
