为了提高透射电镜对组织观察的准确性,在进行电镜样品超薄切片前,对大多数研究样品以及临床外检的标本,需做半薄切片进行光镜定位。为缩短整个定位程序的时间,提高电镜观察效率,我们以Field氏染法为基础,改进了半薄切片染色方法,应用于国产环氧树脂618包埋组织的半薄切片染色。通过对肾、肺、胃、皮肤、十二指肠、支气管、骨、神经、耳蜗、角膜等组织染色,均获得较满意的效果。与
In order to improve the accuracy of TEM observation, we need to make semi-thin sections for most of the study samples and the specimens for clinical examination before performing ultra-thin sectioning of SEM. In order to shorten the time of the whole positioning process and improve the efficiency of electron microscope observation, we improved the semi-thin section dyeing method based on the Field’s staining method and applied it to the semi-thin section staining of the domestic epoxy 618 embedded tissue. Through the kidney, lung, stomach, skin, duodenum, bronchus, bone, nerve, cochlea, cornea and other tissue staining, are more satisfactory results. versus
