目的:探讨中药复方水溶液体系下黄连解毒汤中主要药效成分与树脂吸附和筛分原理之间的内在关联,为黄连解毒汤的剂型改进奠定实验基础。方法:选择目前市售的11种不同类型大孔树脂进行静态吸附和解析实验。RP-HPLC比较各组水溶液体系精制前后生物碱、黄酮、环烯醚萜苷三大类药效中指标性成分含量的动态变化。结果:不同类型大孔树脂对黄连解毒汤中三大类药效成分的吸附率、解析率差异均较大。极性较小、平均孔径较大的X-5,AB-8型树脂对生物碱类的吸附率、解析率均较高;表面积和平均孔径较大的X-5,ADS-17,ADS-7,NKA-9,D151型树脂对黄酮类成分的富集作用较好;平均孔径较大的X-5,S-8,NKA-Ⅱ型树脂和极性较大的ADS-7,D151型树脂对环烯醚萜苷类成分的富集作用较好高。结论:在中药复方水溶液体系下,采用树脂组合的方式,可以优化目标产物的分离工艺。
OBJECTIVE: To explore the inherent relationship between the main active ingredient in Coptis chinensis decoction and the principle of resin adsorption and screening under the traditional Chinese medicine compound aqueous solution system, and lay an experimental foundation for the improvement of the dosage form of Huanglian Jiedu Decoction. Methods: Eleven different types of macroporous resins commercially available were selected for static adsorption and analytical experiments. RP-HPLC was used to compare the dynamic changes of the index components in the three major classes of alkaloids, flavones and iridoid glycosides before and after purification. Results: Different types of macroporous resin Huanglian Jiedu Decoction of the three major types of active ingredient adsorption rate, resolution were large differences. X-5 and AB-8 resins with smaller polarities and larger average pore diameters have higher adsorption rates and resolution rates for alkaloids; X-5, ADS-17 and ADS- 7, NKA-9, D151 type resin better flavonoid enrichment; X-5, S-8, NKA-Ⅱ type resin with larger average pore size and the more polar ADS-7, D151 type Resin on the iridoid glycoside glycoside composition enrichment role is high. Conclusion: Under the traditional Chinese medicine compound aqueous solution system, the resin combination method can be used to optimize the separation process of the target product.
