利用DNA条形码技术对蜈蚣药材进行鉴别,为蜈蚣药材鉴定提供新的方法。该研究以COI条形码序列为基础,对蜈蚣实验样品进行DNA提取,PCR扩增和双向测序,用MEGA6.0软件对所有5个物种的50份样品进行序列比对和邻接(NJ)树构建。结果显示实验样品均可以获得COI序列,蜈蚣药材与其混伪品COI序列种间平均K2P距离为0.222,种间最小K2P距离为0.190,基于COI序列构建的邻接(NJ)树中少棘巨蜈蚣单独聚在一枝,与其混伪品可以相互区分。因此基于COI条形码序列可以准确鉴定蜈蚣及其混伪品。
Identification of centipede medicinal materials by DNA barcoding technology provides a new method for the identification of centipede medicinal materials. In this study, centipede samples were extracted with DNA, PCR amplification and two-dimensional sequencing based on the COI barcode sequence. MEGA 6.0 software was used to construct the sequence alignment and neighbor-joining (NJ) of 50 samples of all 5 species. The results showed that the COI sequences were obtained in all the experimental samples. The average K2P distance between centipede medicinal plants and their counterfeit COI sequences was 0.222, and the minimum distance between species was 0.190. Based on the COI sequence, the spikelet centipede Gather in a branch, with their adulterated products can be distinguished from each other. Therefore, centipede and its adulteration products can be accurately identified based on the COI barcode sequence.
