云南铜壁关省级自然保护区发现红鬣羚

红鬣羚Capricornis rubidus隶属鲸偶蹄目Cetartiodactyla牛科Bovidae鬣羚属(魏辅文等,2021 ) ,被《濒危野生动植物种国际贸易公约(CITES)》列入附录Ⅰ(CITES,2019),被《世界自然保护联盟( IUCN)濒危物种红色名录》评估为近危(NT)(Duckworth & Than, 2008).2017年之前,红鬣羚仅知分布于缅甸北部(Grubb, 2005),其分布区的东北部与中国云南省傈僳族自治州接壤,但中国境内一直未有确认的野外记录.《中国动物地理》(张荣祖,1999 )、《中国哺乳动物多样性及地理分布》(蒋志刚等,2015 )和《中国脊椎动物红色名录》(蒋志刚等, 2016 )均未记录红鬣羚.2014年,红外相机先后在云南省的腾冲市( Li et al.,2019;表1)和怒江傈僳族自治州泸水县(Chen et al.,2019;表1)拍摄到红鬣羚,并被作为中国新记录种正式报道( Li et al., 2019).2021年红鬣羚被列为国家二级重点保护野生动物(国家林业和草原局,农业农村部,2021).

林麝Moschus berezovskii是国家一级重点保护野生动物.本研究从林麝基因组和已发表的微卫星位点中筛选到7个适合林麝粪便DNA扩增的四碱基微卫星位点,建立了林麝的个体识别体系.在四川白水河国家级自然保护区20条监测样线中共收集到林麝粪便46份,基于微卫星个体识别鉴定到30只个体,使用微卫星和线粒体D-Loop进行多样性评估,结果显示,7个微卫星位点的平均期望杂合度和观察杂合度分别为0.602和0.563,4个位点显著偏离哈迪-温伯格平衡,且均表现为杂合不足.线粒体D-Loop遗传多样性检测结果
江西武夷山国家级自然保护区内分布的琴蛙属Nidirana物种此前均被记录为弹琴蛙N.adenopleura.通过对该保护区及周边地区的琴蛙种群进行采样,结合形态比较和线粒体COⅠ基因的分子系统发育分析,发现保护区及周边地区的琴蛙种群包含2个不同的物种——弹琴蛙和孟闻琴蛙N.mangveni.孟闻琴蛙系江西省两栖类新记录,这一发现将孟闻琴蛙的分布区域由已知的浙江北部和东部向南扩展至赣东北地区,为琴蛙属的系统学和谱系地理学研究提供了重要信息.此外,讨论了琴蛙属内物种同域分布的现象,并提出需要对保护区及周边地区
CRISPR/Cas9系统介导的定点突变已经在多种模式生物中实现,其中提高sgRNA的编辑效率是关键性因素.通过分析斑马鱼Danio rerio sgRNA序列特征,优化现有sgRNA设计原则,提高了CRISPR/Cas9系统在斑马鱼中的切割效率.根据现有的sgRNA设计原则,利用软件CHOPCHOP和Benchling对斑马鱼内35个基因设计157个sgRNAs并分析其序列特征对基因突变效率的影响.结果表明,CRISPR/Cas9系统对斑马鱼产生高效靶向突变的sgRNA序列具有明显的碱基偏好性.当靶点序
