目的 探讨白细胞介素 (IL) 3、IL 6和IL 8在脑膜瘤细胞中的表达和作用 ,以及体外针对这种生长调节环路抑制脑膜瘤细胞增殖的可能性。方法 原代培养起 2 1例脑膜瘤细胞。采用ABC免疫细胞化学法明确IL 3、IL 6和IL 8在脑膜瘤细胞中的表达 ,并选用四唑盐 (MTT)比色法观察上述细胞因子及IL 3单抗对脑膜瘤细胞增殖的调控作用。结果 脑膜瘤细胞可分泌IL 3、IL 6和IL 8。IL 3和IL 6皆以剂量依赖关系刺激培养脑膜瘤细胞增殖 ,而IL 8仅在 1 0ng/ml时对脑膜瘤细胞增殖的刺激效应有统计学差异。IL 3单抗以剂量依赖关系分别抑制正常脑膜瘤细胞及 50 %V/V脑膜瘤培养提取液刺激的脑膜瘤细胞增殖。结论 IL 3、IL 6和IL 8为脑膜瘤自泌细胞因子 ;IL 3和IL 6刺激体外培养脑膜瘤细胞增殖 ;IL 3单抗抑制体外培养脑膜瘤细胞增殖 ,为脑膜瘤的生物治疗提供新的理论基础。
Objective To investigate the expression and the role of interleukin (IL) 3, IL 6 and IL 8 in meningioma cells and the possibility of inhibiting the proliferation of meningioma cells in this growth-regulating loop in vitro. Methods Primary culture of 21 cases of meningioma cells. The expression of IL-3, IL-6 and IL-8 in meningioma cells was determined by ABC immunocytochemistry. The cytokines and IL-3 monoclonal antibodies were used to detect the proliferation of meningioma cells by MTT assay effect. Results The meningioma cells secreted IL-3, IL-6 and IL-8. Both IL-3 and IL-6 stimulated the proliferation of meningioma cells in a dose-dependent manner. However, IL-8 stimulated the proliferation of meningioma cells only at 10ng / ml. IL-3 monoclonal antibody inhibited the proliferation of meningioma cells stimulated by culture extract of normal meningioma cells and 50% V / V meningiomas in a dose-dependent manner. Conclusions IL 3, IL 6 and IL 8 are meningioma autologous proinflammatory cytokines; IL 3 and IL 6 stimulate the proliferation of meningioma cells cultured in vitro; IL 3 monoclonal antibody inhibits the proliferation of meningioma cultured in vitro and provides new biological therapy for meningiomas The theoretical basis.
