目的:建立HPLC法测定芝麻油中芝麻素、芝麻林素含量的方法。方法:以芝麻为原料,通过索式提取,皂化预处理,经石油醚提取、浓缩,甲醇定容,利用高效液相色谱测定不同芝麻原料对应产品中芝麻素、芝麻林素的含量范围。结果:被测两峰完全分离,且峰形较好,线性范围10~100μg/m l,相关系数rsesm in=0.999619;rsesam olin=0.999723。芝麻素、芝麻林素平均回收率分别为102.1%,100.2%。精密度分别为芝麻素的RSD=2.31%;芝麻林素的RSD=3.05%。最低检出限:芝麻素为1.7μg/m l,芝麻林素为2.5μg/m l(以进量浓度计);对应产品中芝麻素的含量范围为2.5~7.0 mg/g,芝麻林素的含量范围为2.5~4.5 mg/g。结论:该方法简单、准确、灵敏度高、可用于芝麻素、芝麻林素的测定,实验结果可以作为鉴别芝麻油真伪的一种判断依据。
Objective: To establish a HPLC method for the determination of sesamin and sesamin in sesame oil. Methods: The content of sesamin and sesaminin in different sesame raw materials was determined by using Sesame as raw material, Soxhlet extraction, saponification pretreatment, petroleum ether extraction, concentration and methanol concentration. Results: The two peaks were completely separated and the peak shape was good. The linear range was 10 ~ 100μg / ml, the correlation coefficient was 0.999619, and rsesam olin = 0.999723. The average recoveries of sesamin and sesamin are 102.1% and 100.2% respectively. The RSD of sesamin was 2.31% and the RSD of sesamin was 3.05%. The detection limit of sesamin was 1.7μg / ml and that of sesamin was 2.5μg / ml. The content of sesamin in the corresponding products ranged from 2.5 mg / kg to 7.0 mg / g. The content of sesaminin The range is 2.5-4.5 mg / g. Conclusion: The method is simple, accurate and sensitive. It can be used for the determination of sesamin and sesamin. The experimental results can be used as a basis for judging the authenticity of sesame oil.
