目的观察糖尿病豚鼠膀胱Cajal样间质细胞形态学变化,探讨糖尿病膀胱功能改变原因。方法用链脲佐菌素单次注射建立豚鼠糖尿病模型,建模成功4周后,行尿动力学检查,采用组织冷冻切片、酪氨酸蛋白激酶受体单克隆抗体间接免疫荧光检测及透射电镜技术观察膀胱中Cajal样间质细胞变化。结果与对照组相比,糖尿病豚鼠膀胱最大逼尿肌压显著降低,最大膀胱容量及膀胱顺应性显著明显增高。免疫荧光检测显示逼尿肌中KIT阳性细胞与逼尿肌组织面积百分比均明显小于对照组。电镜下可见Cajal样间质细胞内细胞器变性、减少,胞质广泛溶解,核周间隙增宽,与其他细胞间的缝隙连接显著减少、结构破坏、连接松散。结论糖尿病豚鼠膀胱Cajal样间质细胞的数量减少和结构破坏可能是造成膀胱功能障碍的重要原因之一。
Objective To observe the morphological changes of Cajal-like interstitial cells in the bladder of diabetic guinea pigs and to explore the reasons of the changes of bladder function in diabetic rats. Methods A guinea pig model of diabetes was established by a single injection of streptozotocin. After 4 weeks of successful model establishment, urodynamic examination was performed. Tissue sections were frozen, indirect immunofluorescence assay of tyrosine kinase receptor monoclonal antibody and transmission electron microscopy The changes of Cajal-like interstitial cells in the bladder were observed. Results Compared with the control group, the maximal detrusor pressure of bladder in diabetic guinea pigs was significantly decreased, and the maximum bladder capacity and bladder compliance were significantly higher. Immunofluorescence showed that the percentages of KIT positive cells and detrusor tissue area in detrusor were significantly lower than those in control group. Electron microscopy showed Cajal-like interstitial cells within the organelles degeneration, decrease, widespread dissolution of cytoplasm, perinuclear gap widened, and other cell gap was significantly reduced connection, structural damage, loose connection. Conclusions The decrease of Cajal-like interstitial cells and the structural damage of bladder in diabetic guinea pigs may be one of the important causes of bladder dysfunction.
