目的:探讨凯力康对大鼠心肌缺血再灌注(I/R)损伤的保护作用及作用机理。方法:SD大鼠50只随机分为假手术组、模型组、凯力康15×10~(-3)PAN U/Kg组、凯力康30x10~(-3)PAN U/Kg组及硝酸山梨酯组(1.0 mg/kg),每组10只。大鼠麻醉后测量左心室内压,记录体表心电图;随后开胸结扎左前降支冠状动脉,造成缺血后30 min再灌注120 min(假手术组除外)。结扎后各组舌下静脉给药,记录缺血前、缺血后及灌注后各项血流动力学指标;测定血清中乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK-MB)、超氧化物歧化酶(SOD)、丙二醛(MDA);计算心脏指数及心肌梗死范围。结果:与模型组相比,凯力康对I,/R大鼠的血流动力学指标均有明显的改善作用;能缩小心肌梗死范围;降低血清中LDH、CK、MDA水平、升高血清中SOD活性。结论:凯力康通过提高血清中SOD含量、降低MAD、LDH、CK含量而对大鼠心肌I/R损伤有明显的保护作用。
Objective: To investigate the protective effect and mechanism of Keli Kang on myocardial ischemia-reperfusion (I / R) injury in rats. Methods: Fifty SD rats were randomly divided into sham operation group, model group, Kaili Kang 15 × 10 ~ (-3) PAN U / Kg group, Kaili Kang 30 × 10 ~ (-3) PAN U / Kg group and nitric acid Sorbitol group (1.0 mg / kg), 10 rats in each group. Left ventricular pressure was measured after anesthesia in rats, and the body surface electrocardiogram (ECG) was recorded. Then the left anterior descending coronary artery was ligated by thoracotomy and the animals were reperfused for 120 min at 30 min after ischemia (except sham group). After ligation, the sublingual vein was administered to each group, and the hemodynamic indexes were recorded before ischemia, after ischemia and after perfusion. The levels of serum LDH, CK-MB, Superoxide dismutase (SOD), malondialdehyde (MDA); Calculated heart index and myocardial infarction range. Results: Compared with the model group, Kailikang could significantly improve the hemodynamic parameters of I / R rats, reduce the range of myocardial infarction, reduce the level of serum LDH, CK and MDA, In the SOD activity. Conclusions: Kelixan has a significant protective effect on myocardial I / R injury in rats by increasing the content of SOD and decreasing the levels of MAD, LDH and CK in serum.
