以大葱(Allium fistulosum L.)雄性不育系和保持系为试材,开发了一个能够鉴别细胞质雄性不育类型的SCAR标记。此标记在大葱雄性不育系(S型细胞质)中扩增出1条607bp片段,而在保持系(N型细胞质)中没有扩增出此片段,将此片段命名为S607。对5组不同遗传背景的不育系和相应保持系,以及4份杂交组合进行验证,SCAR标记S607鉴定结果与实际细胞质类型完全相符。
A SCAR marker capable of identifying cytoplasmic male sterility was developed using the male sterile line and maintainer line of Allium fistulosum L. as test material. This marker amplified a 607bp fragment in Welsh onion CMS line (S-type cytoplasm), while it was not amplified in maintainer line (N-type cytoplasm). The fragment was named as S607. Five groups of sterile lines with different genetic backgrounds and corresponding maintainer lines, as well as four crosses, were validated. The result of SCAR marker S607 was completely consistent with the actual cytoplasm type.
