目的探讨胰岛素样生长因子ⅡmRNA结合蛋白IMP1在食管癌和贲门癌及癌前病变组织的表达变化特征及其生物学意义。方法采用免疫组化ABC法检测123例食管和贲门癌及癌前病变组织IMP1蛋白的表达状况和特征。结果食管:IMP1在正常鳞状上皮组织表达最高(100%),并随病变进展,阳性表达率呈下降趋势,正常食管上皮与基底细胞过度增生(BCH,91·67%)、不典型增生(DYS,85·71%)和鳞状细胞癌(SCC,83·33%)之间差有异显著性(P<0·05),且BCH与SCC之间阳性表达率差异明显(P<0·05)。贲门:IMP1在各级病变组织均有不同程度表达,不典型增生组织(DYS,83·33%%)阳性率最高,表达最低是慢性萎缩性贲门炎组织(CAG,54·55%),DYS与正常贲门腺上皮(61·11%)、CAG和贲门腺癌(GCA,61·90%)之间差异有显著性(P<0·01)。IMP1表达率与食管鳞癌和贲门腺癌分化程度无相关性(P>0·05)。结论IMP1蛋白表达变化是食管癌、贲门癌和癌前病变组织常见的分子事件,可能在食管鳞癌和贲门腺癌癌变过程中起一定作用。
Objective To investigate the expression and significance of insulin-like growth factor Ⅱ mRNA binding protein IMP1 in esophageal and gastric cardia carcinoma and precancerous lesions. Methods Immunohistochemical ABC method was used to detect the expression and characteristics of IMP1 protein in 123 esophageal and gastric cardia cancers and precancerous lesions. RESULTS: The expression of IMP1 in normal squamous epithelium was the highest (100%). With the progression of the disease, the positive expression rate of IMP1 showed a decreasing trend. The normal esophageal epithelium and basal cell hyperplasia (BCH, 91.67%), dysplasia DYS, 85.71%) and squamous cell carcinoma (SCC, 83.33%) were significantly different (P <0.05), and the difference between BCH and SCC was significant (P <0 · 05). Cardia: The expression of IMP1 in all stages of the lesion was different. The positive rate of dysplasia (DYS, 83.33%) was the highest, and the lowest was IMP of CAG (54.55%), DYS There was a significant difference between normal gastric cardia epithelium (61.11%), CAG and GCA (61.90%) (P <0.01). The expression of IMP1 had no correlation with the differentiation of esophageal squamous cell carcinoma and gastric cardia adenocarcinoma (P> 0.05). Conclusion The expression of IMP1 protein is a common molecular event in esophageal cancer, cardia cancer and precancerous lesions, which may play a role in the carcinogenesis of esophageal squamous cell carcinoma and gastric cardia adenocarcinoma.
