膝痹膏摩法对膝痹兔滑膜基质金属蛋白酶的影响

目的:探讨膝痹膏摩法对膝痹兔滑膜基质金属蛋白酶的影响。方法:66只健康的新西兰大白兔,随机分10只为正常组对照,剩余用石膏铁丝固定法复制膝痹动物模型,8周时在模型中随机处死6只动物查看造模情况,造模成功后将其随机分模型组、手法组和膝痹膏高浓度、中浓度、低浓度组,膝痹膏3个浓度组给予膝痹膏摩法干预,手法组给予单纯手法,空白组和模型组不干预;干预8周处死动物,取滑膜利用ELISA试剂盒检测MMP1、MMP3、MMP13表达。结果:病理检测观察膝痹膏摩法不同浓度组均可改善关节软骨组织学结构,模型组膝关节软骨出现明显的关节软骨损伤。模型组中MMP-1、MMP-3、MMP-13含量均高于其余5组(P<0.05),正常组中MMP-1、MMP-3、MMP-13含量最低(P<0.05)。各组间比较:膝痹膏中浓度组MMP-1、MMP-3的含量明显低于高浓度、低浓度和手法组(P<0.05),而高浓度、低浓度和手法组之间比较没有统计学意义;膝痹膏中浓度组MMP-13低于低浓度和手法组(P<0.05),但与高浓度比较没有统计学意义。结论:膝痹膏摩法可明显改善膝痹的病理表现,其作用机制可能与3个金属蛋白酶的含量降低有关,进而降低了软骨基质的降解,缓解膝痹的进程,仍需进一步研究其具体的作用靶点,完善其作用机制。 Objective: To investigate the effect of Kubi cream on synovial matrix metalloproteinase (MMP) in knee palsy. Methods: Sixty-six healthy New Zealand white rabbits were randomly divided into 10 normal control groups. The rest were treated with gypsum wire fixation to copy the knee joint animal model. At 8 weeks, 6 animals were randomly sacrificed in the model to check the model making. The rats were randomly divided into model group, manipulation group and Kubi Gao high concentration, medium concentration, low concentration group and Kubi Ointment group. The three concentration groups were given Kubi Māmori intervention. The manipulation group was given simple manipulation, blank group and model group No intervention; Animals were sacrificed 8 weeks after intervention, and the synovial membrane was used to detect the expression of MMP1, MMP3 and MMP13 by ELISA kit. Results: The pathological examination showed that the different concentrations of Kubi Momo method could improve the histological structure of articular cartilage, and the articular cartilage of articular cartilage of model group showed obvious injury. The levels of MMP-1, MMP-3 and MMP-13 in the model group were significantly higher than those in the other 5 groups (P <0.05). The content of MMP-1, MMP-3 and MMP-13 in the normal group was the lowest (P <0.05). The comparison among groups showed that the contents of MMP-1 and MMP-3 in Kubi Oral Concentration group were significantly lower than those in high concentration, low concentration and manipulation groups (P <0.05), while those in high concentration, low concentration and manipulation groups were not Statistical significance; Kubi concentration in the concentration group MMP-13 was lower than the low concentration and manipulation group (P <0.05), but compared with the high concentration was not statistically significant. Conclusion: Kubi Moxibustion can significantly improve the pathological manifestations of knee pain, the mechanism may be related to the reduction of three metalloproteinases, thereby reducing the degradation of cartilage matrix, to alleviate the knee process, the need for further study of its specific The role of target, improve its mechanism of action.
