背景:鞘氨醇1磷酸受体(sphingosine 1~-phosphate(S1P)receptor,S1PR)是淋巴细胞表面一种G~-蛋白偶联受体,其配体是S1P。S1P/S1PR间的相互作用对淋巴细胞在体内的分布和迁移具有重要影响。目的:探讨S1PR5基因缺陷对体内淋巴细胞分布的影响。方法:用流式细胞术检测S1PR5基因缺陷对小鼠各组织器官中淋巴细胞亚群分布的影响。结果:与野生型小鼠相比,S1PR5缺陷小鼠的外周血(PB)和脾脏(SP)中NK细胞的比例降低(PB:6.4±0.45%vs 2.2±0.47%)(P<0.01,n=3);(SP:3.0±0.91%vs 0.68±0.14%)(P<0.05,n=3),而骨髓(BM)和淋巴结(LN)中NK细胞的比例升高(BM:0.97±0.20%vs 2.6±0.35%)(P<0.01,n=3);(LN:0.35±0.16%vs 1.7±0.15%)(P<0.01,n=3),而CD3~+T淋巴细胞在外周血、脾脏和淋巴结中的比例则无明显变化(PB:17.3±7.9%vs 17.0±4.6%)(P>0.05,n=3);(SP:33.0±6.0%vs 27.4±1.8%)(P>0.05,n=3);(LN:42.3±10.7%vs 51.2±2.7%)(P>0.05,n=3)。结论:S1PR5缺陷对小鼠体内NK细胞的分布产生重要影响。
BACKGROUND: Sphingosine 1 -phosphate (S1P) receptor, S1PR) is a G ~ - protein coupled receptor on the surface of lymphocytes. Its ligand is S1P. The interaction between S1P / S1PR has important implications for the distribution and migration of lymphocytes in vivo. Objective: To investigate the influence of S1PR5 gene on the distribution of lymphocytes in vivo. Methods: The effect of S1PR5 gene defect on the distribution of lymphocyte subsets in various tissues and organs of mice was detected by flow cytometry. Results: The proportion of NK cells in peripheral blood (PB) and spleen (SP) of S1PR5-deficient mice was decreased (PB: 6.4 ± 0.45% vs 2.2 ± 0.47% (P <0.05, n = 3), while the proportion of NK cells in bone marrow (BM) and lymph node (LN) increased (BM: 0.97 ± 0.20 % vs 2.6 ± 0.35%) (P <0.01, n = 3); LN (0.35 ± 0.16% vs 1.7 ± 0.15% (P> 0.05, n = 3); (SP: 33.0 ± 6.0% vs 27.4 ± 1.8%, P> 0.05), but there was no significant difference in the proportion of spleen and lymph nodes (PB: 17.3 ± 7.9% vs 17.0 ± 4.6% 0.05, n = 3); (LN: 42.3 ± 10.7% vs 51.2 ± 2.7%) (P> 0.05, n = 3). Conclusion: S1PR5 deficiency has an important effect on the distribution of NK cells in mice.
