目的 建立同时测定贝母中 5种异甾类生物碱———peimissine ,imperialine ,sinpeinineA ,imperialine 3β glucoside和yibeinosideA含量的HPLC分析方法。方法 C1 8柱 ;流动相 :乙腈 水 (含 0 1%二乙胺 ) ;梯度洗脱 ,流速 1 0mL·min- 1 ;检测器 :Alltech 5 0 0蒸发光散射检测器 (ELSD)。结果 线性范围为peimissine 13 1~ 2 88 2mg·L- 1 (r2 =0 9975 ) ,imperialine 3β glucoside 7 7~ 16 9 4 mg·L- 1 (r2 =0 9993) ,yibeinosideA 7 3~ 16 0 6mg·L- 1 (r2 =0 9997) ,imperialine 16 5~ 36 3 0mg·L- 1 (r2 =0 9992 ) ,sinpeinineA 8 7~ 191 4mg·L- 1 (r2 =0 994 2 )。5个化合物的精密度和重现性RSD均 <5 %。结论 本方法简便、有效、可行 ,可用于贝母中 5种异甾类生物碱的含量测定。
OBJECTIVE To establish an HPLC method for simultaneous determination of five isosteroid alkaloids, peimissine, imperialine, sinpeinine A, imperialine 3β glucoside and yibeinosideA in Fritillaria thunbergii. Method C1 8 column; mobile phase: acetonitrile water (containing 0.1% diethylamine); gradient elution at 10 mL · min -1; detector: Alltech 500 evaporative light scattering detector (ELSD). The linear range of the results was peimissine 13 1 ~2 88 2 mg · L -1 (r2 = 0 9975), imperialine 3β glucoside 7 7 ~ 16 9 4 mg · L- 1 (r2 = 0 9993), yibeinoside A 7 3 ~16 0 6 mg · L 1 (r2 = 0 9997), imperialine 16 5 ~ 36 3 0 mg · L -1 (r 2 = 0 9992), sinpeinine A 8 7 ~ 191 4 mg · L -1 (r 2 = 0 994 2). The RSDs were <5% for the five compounds for both precision and reproducibility. Conclusion The method is simple, effective and feasible, and can be used for the determination of five isosterol alkaloids in Fritillaria.
