目的确定鳖甲多糖的最佳提取工艺,以苯酚硫酸法测定鳖甲多糖含量。方法采用碱提法提取、醇沉得到鳖甲多糖;利用苯酚硫酸法显色,在490nm处测定吸光度。结果碱液浓度为8%、料液比为1∶25、提取温度为60℃、提取时间8h为最佳提取工艺,最佳醇沉浓度为80%;平均回收率为100.2%,RSD为1.4%(n=9)。结论本实验鳖甲多糖提取及含量测定操作简便、结果准确、重复性好,可用于鳖甲多糖的提取与含量测定。
Objective To determine the best extraction technology of turtle polysaccharides, and to determine the polysaccharide content of turtle by phenol - sulfuric acid method. Methods Alkaloid extraction and alcohol precipitation were used to obtain the polysaccharides from turtle shell. The phenolics sulfuric acid method was used to determine the absorbance at 490 nm. Results The optimum extraction conditions were as follows: the concentration of lye was 8%, the ratio of solid to liquid was 1:25, the extraction temperature was 60 ℃, the extraction time was 8h, the optimum alcohol concentration was 80%; the average recovery was 100.2% and the RSD was 1.4 % (n = 9). Conclusion The extraction and determination of polysaccharide from turtle shell in this experiment is simple, accurate, reproducible and can be used for extraction and determination of polysaccharide from turtle shell.
