为了研究奶牛黄体的形成及退化过程的相关机理,并进一步探究更加准确的妊娠及相关生理过程的蛋白标记物,试验采用蛋白质组学经典的二维凝胶电泳(2-DE)技术检测和分析妊娠前后黄体组织间的差异蛋白,使用PDQuest8.0软件检测,取差异蛋白点进行分析,并经MALDI TOF/TOFMS/MS检测。结果表明:6种差异蛋白分别为26s蛋白酶体蛋白、纤维蛋白、DNA拓扑异构酶蛋白、核纤层蛋白、硫氧还原蛋白、微管蛋白。对这6种蛋白在组织间的表达差异进行分析,发现这些蛋白可能与黄体形成和黄体退化及黄体激素分泌有关。
In order to study the mechanism of luteal formation and degradation in dairy cattle, and to further explore more accurate protein markers of pregnancy and related physiological processes, the experiments were performed using proteomic classic 2-dimensional gel electrophoresis (2-DE) The difference protein between luteal tissues before and after pregnancy was detected by PDQuest8.0 software, the difference protein spots were analyzed and detected by MALDI TOF / TOF MS / MS. The results showed that six kinds of differential proteins were 26s proteasome protein, fibrin, DNA topoisomerase, lamin, thioredoxin and tubulin respectively. The differences in expression of these six proteins between tissues were analyzed and found that these proteins may be associated with corpus luteum formation and corpus luteum degeneration and progesterone secretion.
